Antibodies to TRAIL Protein Family - FocusOn 022
The TNF-related apoptosis-inducing ligand (TRAIL, TNFSF10) is a member of the TNF superfamily. The interaction of TRAIL with its receptors (see below) either induces or blocks apoptosis.
The TRAIL ligands TRAIL-R1 (DR4, CD261), TRAIL-R2 (Apo2, DR5, CD262, TRICK2) and TRAMP (DR3, Apo-3, WSL-1, LARD, DDR3, TR3) activate NF-kappaB, and induce TRAIL-mediated apoptosis. Initially, TRAMP (gene name: TNFRSF25) was thought to be a receptor for TWEAK (see Focus on No. 18 - Antibodies to TWEAK and TWEAK Receptor), but further studies have shown that TWEAK could induce apoptosis via receptors distinct from TRAMP.
TRAIL-R3 (DcR1, CD263, TRID, LIT) is a glycophospholipid-anchored cell surface protein. It has an extracellular TRAIL binding domain and lacks the cytoplasmic death domain (DD) shared by several members of the TNF receptor superfamily. TRAIL-R4 (DcR2, CD264, TRUNDD) is a transmembrane cell surface protein. It has an extracellular TRAIL binding domain and a truncation in the DD of its cytoplasmic tail. TRAIL-R3 (gene name: TNFRSF10C) and TRAIL-R4 (gene name: TNFRSF10D) protect cells from TRAIL-mediated cytotoxicity: They bind to TRAIL and inhibit binding of TRAIL to TRAIL-R1 (gene name: TNFRSF10A and TRAIL-R2 (gene name: TNFRSF10B).
