| Add. information | Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies reacting with other dog serum proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum. |
| Application | Can be used in precipitating techniques as Immunoelectrophoresis and single or Double Radial Immunodiffusion to identify the presence of C5 containing complexes in human plasma or other body fluids or to determine its concentration. The presence of non precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in more sensitive non-precipitating antibody-binding techniques if proper controls are included. Recommended Dilutions: Immunoelectrophoresis: Use 2 µl human plasma or equivalent against 120 µl antiserum. Double Radial Immunodiffusion: Use a rosette arrangement with 10 µl antiserum in 3 mm diameter center well and 2 µl plasma samples (neat and serially diluted) in 2 mm diameter peripheral wells. Antibody Titre: Precipitin titre 1/32 when tested against in agar-block immunodiffusion titration. |
| Background | C5 component of human complement is a plasma protein consisting of two polypeptide chains, alpha, beta, bound by disulphide bonds. It has a molecular weight of 190,000. The activated protein forms with the complement factors C6-C9 the membrane attack complex. The average concentration in plasma is about 70 µg/ml. |
| Concentration | Total protein and IgG concentrations in the antiserum are comparable to those of normal goat serum. No foreign proteins added. |
| Storage | Store lyophilized at 2-8°C and reconstituted at 2-8°C for one month or (in aliquots) at -20°C for longer. Avoid Repeated thawing and freezing. Shelf life: one year from despatch |
