| Application | Intended use In precipitating techniques as Immunoelectrophoresis and single and double radial Immunodiffusion to identify the presence Transferrin in Swine serum or other body fluids and to determine its concentration. This antiserum has not been tested in more sensitive assays as nephelometry, ELISA or use in immunohistology, but this does not exclude such use if proper controls are included. Recommended Dilutions: Immunoelectrophoresis: Use 2 µl or equivalent against 120 µl antiserum. Double Radial Immunodiffusion (Ouchterlony): Use a rosette arrangement with 10 µl in a 3 mm diameter center well and 2 µl serum samples (neat and serially diluted) in 2 mm diameter peripheral wells. Precipitin titre: 1/32 when tested against pooled normal Swine serum in agar-block Immunodiffusion titration. |
| Background | Transferrin is a single polypeptide chain glycoprotein belonging to iron binding family of proteins. It has a molecular weight of 81,5 kDa (porcine). It is synthesised in the liver and consists of two domains each having a high affinity reversible binding site for Fe3+. The iron is transported in blood and interstitial fluids to sites of use and disposal. Iron/transferrin is essential in haemoglobin synthesis and for certain types of cell division. Serum concentration rises in iron deficiency and pregnancy and falls in iron overload, infection and inflammatory conditions. |
| Concentration | Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added. |
| Storage | Store lyophilized at 2-8°C and reconstituted at 2-8°C for one month or (in aliquots) at -20°C for longer. Avoid Repeated thawing and freezing. Shelf life: one year from despatch. |
