Immunogen
Immunogen:
Highly purified normal IgG isolated from pooled rhesus monkey serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Add. information Adsorption: Immunoaffinity adsorbed using insolubilized antigens as required, to eliminate antibodies cross-reacting with other components of the immunoglobulin system or reacting with other serum proteins. The use of insolubilized adsorption antigens prevents the presence of excess adsorbent protein or immune complexes in the antiserum.
Application Can be used in precipitating techniques as immunoelectrophoresis and radial immunodiffusion to identify the presence of IgG in monkey serum or other body fluids or to determine its concentration. To prepare an immunoadsorbent for the purification of monkey IgG from serum or plasma.
Directions for use:
In immunoelectrophoresis use 2 μl serum or equivalent against 120 μl antiserum.
In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 μl antiserum in 3 mm diameter center well and 2 μl serum samples (neat and serially diluted in 2 mm diameter peripheral wells.
Storage Prior to reconstitution store at 2-8°C.
Following reconstitution store undiluted at 2-8°C for one week
or (in aliquots) at -20°C for longer.
Avoid repeated freezing and thawing.
Shelf life: one year from despatch.

Specificity
Specificity:
The reactivity of the antiserum is restricted to the Fc part of the IgG molecule. In immunoelectrophoresis and radial immunodiffusion, using various antiserum concentrations against normal monkey serum a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified IgG. No precipitation reaction is obtained with purified IgA, IgM, and IgG/Fab fragments.
Cross-reactivity: Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Precipitation reactions have been observed with immunoglobulins in serum of other old-world monkeys, including Cercopithecus, Cynomolgus and Baboon. The antiserum may also react with other species as has been observed for chimpanzee and man.
