Immunogen
Immunogen:
Purified normal IgG isolated from pooled rhesus monkey serum. Freund’s complete adjuvant is used in the first step of the immunization procedure.
Add. information Antibody titre: Precipitin titre not less than 1/128 when tested against normal monkey serum in agar block titration.
Application Can be used in precipitating techniques as immunoelectrophoresis and double radial immunodiffusion to identify the presence of IgG; as secondary antibody to precipitate the immunoglobulin in normal monkey serum; to prepare an immunoadsorbent for the production of Ig-free monkey serum, plasma or non-Ig preparations.
Directions for use:
In immunoelectrophoresis use 2 μl or equivalent against 120 μl antiserum.
In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 μl antiserum in a 3 mm diameter centre well and 2 μl serum samples (neat and diluted) in 2 mm diameter peripheral wells.
Storage Prior to reconstitution store at 2-8°C.
Following reconstitution store undiluted at 2-8°C for one week
or (in aliquots) at -20°C for longer.
Avoid repeated freezing and thawing.
Shelf life: one year from despatch.

Specificity
Specificity:
The reactivity of the antiserum is directed to the Fc and Fab subunits of the IgG molecule. It includes a certain degree of reactivity with other immunoglobulins via the common Fab portion. It does not react with any non-Ig protein in monkey serum, as tested by immuno-
electrophoresis and double radial immunodiffusion.
Cross-reactivity:
Inter-species cross-reactivity is a normal feature of antibodies to immunoglobulins, since Ig of different species frequently share antigenic determinants. Precipitation reactions have been observed with IgG in serum of other old-world monkeys, including Cercopithecus, Cynomolgus and Baboon. The antiserum may also react with IgG of other species as has been observed for chimpanzee and man. Other results of cross reactivity testing using double radial immunodiffusion are as follows:
