| Add. information | Cell lysate was prepared by homogenization in ice-cold modified RIPA Lysis Buffer with cocktail of protease inhibitors (Sigma). Cell debris was removed by centrifugation. Protein concentration was determined with Bradford assay (Bio-Rad protein assay, Microplate Standard assay). The lysate was adjusted to 3.75 mg/ml and mixed with 5x SDS sample buffer to become 1xSDS sample buffer (2% SDS, 300 mM beta-mercaptoethanol, 50 mM Tris-HCl pH 6.8, 10 % Glycerol, & 0.01% Bromophenol blue) at final 3 mg/ml. The lysate was heated at 95°C for 5 min, and cooled rapidly. Lysate quality was assured with a 12% SDS-PAGE Gel stained with Commassive brilliant blue. |
