Background of Bcl-2-like 2 antibody
The Bcl-2 family of proteins is characterized by its ability to modulate cell death (apoptosis) under a broad range of physiological conditions. Bcl-2, A1 and Bcl-xL function to inhibit apoptosis while other members of the Bcl-2 family, Bax, Bad, Bak and Bcl-xS oppose their death-suppressing effects. Using a PCR-based strategy, an additional protein with life-promoting activity, designated Bcl-w, has been identified. The protein is highly conserved between mouse and human and is encoded by a gene located near the TCRalpha gene on chromosome 14. Bcl-w is expressed in myeloid cell lines but not in T and B lymphocytes, and can be found in a wide range of tissues. An alternative splicing event in exon 4 results in two transcripts. The first, Bcl-w, encodes a protein of 193 amino acids, and the second, Bcl-w/rox, encodes a protein 333 amino acids in length. The “rox” portion of Bcl-w/rox shows a striking 66% amino acid sequence identity with the Drosophila rox2 protein; however, the Bcl-w/rox transcript may be expressed at very low levels.

IP/Western blot showing the detection of BCL2L2 in human and mouse cell lines. Lysates from 107 cells were immunoprecipitated by clone 13F9 and the fractionated proteins were then blotted with BCL2L2 monoclonal antibody, clone 13F9 ( Cat # MAB3487 ). Hsp-70 expression was used as an equal loading control and 293T cells transiently transfected with a human bcl-w expression construct were used as a positive control.

Immunohistochemical analysis of BCLW staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Immunofluorescent analysis of BCLW staining in HL60 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
