Background of CD8 antibody
The T cell antigen receptor (TCR) recognizes foreign antigens and translates such recognition events into intracellular signals that elicit a change in the cell from a dormant to an activated state. TCR is a heterodimer composed of either α and β or γ and δ chains. The vast majority of circulating T cells (95%) express the α/β heterodimer while roughly 2-5% express the γ/δ heterodimer.
CD3 chains and the CD4 or CD8 coreceptors are also required for efficient signal transduction through the TCR. The TCR is expressed on T helper and T cytotoxic cells that can be distinguished by their expression of CD4 and CD8. T helper cells express CD4 proteins and T cytotoxic cells display CD8. CD4 is also expressed on cortical cells, mature medullary thymocytes, microglial cells and dendritic cells. CD4, also designated T4 and Leu 3, is a 55 kDa membrane glycoprotein that contains four extracellular immunoglobinlike domains. The TCR, in association with CD4, can bind class II MHC molecules presented by the antigen-presenting cells. The CD4 protein functions by increasing the avidity of the interaction between the TCR and an antigen-class II MHC complex.

Western Blot: CD8 alpha Antibody - Western Blot was performed using affinity purified SEQer CD8a antibody, aa(71-170) antibody. The lanes contain 5-30ug of a whole cell extract. Final concentration of antibodies = 0.1ug/ml (1:10,000 dilution). The blot was probed overnight with the SEQer CD8a antibody, aa(71-170) antibody. Blot was then washed according to protocol and probed with goat-anti-Rabbit-HRP conjugate at 1:5000 dilution, washed and developed using chemiluminescence (film exposure 5-30sec). The protein was detected as represented by the band shown.
HEK293T cells were transfected with the pCMV6-ENTRY control (Left lane) or pCMV6-ENTRY CD8A (RC213165, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-CD8A.

Formalin-fixed and paraffin-embedded human lung carcinoma tissue reacted with CD8A antibody (C-term) (Cat.#TA302179), which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.
