Background of CEBPD antibody

C/EBP proteins are DNA-binding proteins that are important transcriptional activators in the regulation of genes involved in immune and inflammatory responses and may play an important role in the regulation of the several genes associated with activation and/or differentiation of macrophages. Members of the C/EBP transcription factor family are related by a high degree of amino acid sequence identity to the basic leucine zipper DNA-binding domain and show distinct but overlapping patterns of tissue- and stage-restricted expression. Osada et al. identified the consensus binding site for the family as RTTGCGYAAY (R = A or G, and Y = C or T). Phosphorylation of C/EBPdelta may increase binding activity, whereas phosphorylation of the alpha and beta family members may decrease binding affinity. C/EBPdelta is a nuclear protein that binds DNA as a dimer and can form stable heterodimers with all other C/EBP family members.

Western blot using Cebpd polyclonal antibody ( Cat # PAB10182 ) shows detection of endogenous protein in whole cell extracts from mouse primary macrophage cells.The blot shows staining as follows : untreated wild-type ( WT ) cells ( lane 1 ), WT cells treated 3 h with LPS ( lane 2 ) and Cebpd knock-out cells treated 3H with LPS ( lane 3 ).The band at ~35 kDa, indicated by the arrowhead, corresponds to endogenous Cebpd.Primary antibody was used at 1 : 15,000.ECL was used for detection.Personal communication, E.Sterneck, NCI, Frederick, MD.

Immunohistochemical analysis of C/EBP delta/epsilon staining in human lung cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

Immunofluorescent analysis of C/EBP delta/epsilon staining in HepG2 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
