Background of Cyclin A1 antibody
The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. The cyclin encoded by this gene was shown to be expressed in testis and brain, as well as in several leukemic cell lines, and is thought to primarily function in the control of the germline meiotic cell cycle. This cyclin binds both CDK2 and CDC2 kinases, which give two distinct kinase activities, one appearing in S phase, the other in G2, and thus regulate separate functions in cell cycle. This cyclin was found to bind to important cell cycle regulators, such as Rb family proteins, transcription factor E2F-1, and the p21 family proteins. Multiple transcript variants encoding different isoforms have been found for this gene.

Proximity Ligation Analysis (PLA) of protein-protein interactions between PTK2 and CCNA1 HeLa cells were stained with anti-PTK2 rabbit purified polyclonal 1:1200 and anti-CCNA1 mouse monoclonal antibody 1:50. Signals were detected by Duolink® 30 Detection Kit 613(red), and nuclei were counterstained with DAPI (blue). Each red dot represents the detection of protein-protein interaction complex.
Immunohistochemical analysis of Cyclin A1/2 staining in human lung cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Immunofluorescent analysis of Cyclin A1/2 staining in NIH3T3 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

Immunohistochemistry: Cyclin A1 Antibody - Most of the normal tissues displayed strong cytoplasmic and/or nuclear positivity. Malignant tissues exhibited moderate cytoplasmic and occasional nuclear immunoreactivity.
