Background of IRAK4 antibody
The Toll-like receptor (TLR) family in mammal comprises a family of transmembrane proteins characterized by multiple copies of leucine rich repeats in the extracellular domain and IL-1 receptor motif in the cytoplasmic domain. Like its counterparts in Drosophila, TLRs signal through adaptor molecules. IL-1 receptor-associated kinases (IRAKs) are important mediators in the signal transduction of Toll/IL-1 receptor (TIR) family members. The cytoplasmic domains of TIR proteins interact with the adapter protein, MyD88. MyD88 then recruits IRAKs (IRAK-1, -2, and M), which in turn interact with other adapter molecules, such as TRAF6 to activate NF-kB and MAPK pathways. Recently, a new member of this family, IRAK-4 has been identified. IRAK-4 may act as an upstream activator of IRAK-1. IRAK-4 is important for LPS activation of TLRS. Mice lacking IRAK-4 are resistant to lethal doses of LPS and are also severely impaired in their responses to viral and bacterial challenges.

IRAK4 polyclonal antibody ( Cat # PAB6731 ) ( 1 µg/ml ) staining of Hela lysate ( 35 µg protein in RIPA buffer ) . Primary incubation was 1 hour. Detected by chemiluminescence.

The 5 x 107 Jurkat cells were stimulated by 50 ng/ml IL-1 for 10 min, collected, lysed, resolved onto 10% SDS-PAGE, transferred onto NC membrane, and followed by an immunoblotting with IRAK4 ( phospho T345 ) monoclonal antibody, clone A8A8 ( Cat # MAB2538 ) at 1 : 500 ( lane 1 ).Lane 1 was pre-incubated with Phosphospecific peptide.Lane 2 was pre-incubate with non-phospho specific peptide.An immunoreactive band is observed around ~51 kDa.
Dot Blot : 1 µg peptide was blot onto NC membrane.A : IRAK4 ( pT345 ). B : IRAK4 ( non phosphorylated ). Followed by IRAK4 ( phospho T345 ) monoclonal antibody, clone A8A8 ( Cat # MAB2538 ) incubation at a 1 : 1000 dilution.
Immunocytochemistry using IRAK4 polyclonal antibody ( Cat # PAB10251 ) shows staining of IRAK4 in HeLa cells.The cells were fixed with paraformaldehyde and reacted with the primary antibody diluted to 1 : 100.
