Background of Junction plakoglobin antibody
The catenins, α, β and γ, are proteins which bind to the highly conserved,intracellular cytoplasmic tail of E-cadherin. Together, the catenin/cadherincomplexes play an important role mediating cellular adhesion. α-catenin was initially described as an E-cadherin associated protein, and since has been shown to associate with other members of the cadherin family, such as N-cadherin and P-cadherin. β-catenin associates with the cytoplasmic portion of E-cadherin, which is necessary for the function of E-cadherin as an adhesion molecule. β-catenin has also been found in complexes with the tumor suppressor protein APC. γ-catenin, also known as plakoglobin, binds with α-catenin and N-cadherin. It has been shown that the transmembrane phosphatase PTPμ associates with catenin/cadherin complexes and may regulate complex signaling.

Proximity Ligation Analysis (PLA) of protein-protein interactions between JUP and APC. Western blot analysis of Jup immunoprecipitated from lysates prepared from A-431 cells treated with pervanadate. The immunoprecipitation was performed using mouse Jup monoclonal antibody. The blots were probed with Jup monoclonal antibody, clone M111 ( Cat # MAB1383 ). Immunohistochemical analysis of Gamma-catenin staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Immunofluorescent analysis of Gamma-catenin staining in A431 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

Formalin-fixed and paraffin-embedded human lung carcinoma reacted with JUP Antibody (C-term), which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated.
