Background of L3MBTL1 antibody

L3MBTL is the homolog of a protein identified in Drosophila as a suppressor of malignant transformation of neuroblasts and ganglion-mother cells in the optic centers of the brain. L3MBTL is localized to condensed chromosomes in mitotic cells. Overexpression of this gene in a glioma cell line results in improper nuclear segregation and cytokinesis producing multinucleated cells.This gene encodes the homolog of a protein identified in Drosophila as a suppressor of malignant transformation of neuroblasts and ganglion-mother cells in the optic centers of the brain. This gene product is localized to condensed chromosomes in mitotic cells. Overexpression of this gene in a glioma cell line results in improper nuclear segregation and cytokinesis producing multinucleated cells. Alternatively spliced transcript variants encoding different isoforms have been identified.

Western blot was performed on HeLa nuclear extract ( HeLa NE ) using L3MBTL polyclonal antibody ( Cat # PAB14105 ) at dilution 1 : 1,000 in TBS-Tween + 5% skimmed milk. The antibody recognizes endogenous L3MBTL1 in HeLa nuclear extract ( HeLa NE ). The location of L3MBTL1 is indicated with an arrow.

Immunoprecipitation was performed on nuclear extracts containing ectopically expressed FLAG-tagged L3MBTL1 ( L3MBTL1-F ) using L3MBTL polyclonal antibody ( Cat # PAB14105 ) and an IgG control antibody ( IgG Control ).The presence of L3MBTL1-F in the input ( In ; Lane 1 ), in the precipitated fraction ( P ) and in the supernatant fraction ( SN ) was tested by Western Blot using anti-FLAG antibody.L3MBTL1-F was efficiently precipitated by L3MBTL polyclonal antibody ( Cat # PAB14105 ) ( Lane 3 ), since there was no L3MBTL1-F present in the supernatant ( Lane 5 ).L3MBTL1-F was not precipitated by the control antibody ( IgG Control ) ( Lane 2 ) and the L3MBTL1 was still present in the supernatant ( Lane 4 ).
