Background of NOS3 antibody
Nitric oxide synthase NOS oxidizes a guanidine nitrogen of arginine releasing nitric oxide in the form of a free radical and citrulline. Nitric oxide thus generated acts as a messenger in diverse functions including vasodilation neurotransmission, anti tumor and anti pathogenic activities. NOS is classified under three types: neuronal NOS (nNOS) or brain NOS (bNOS); inducible NOS (iNOS) or macrophage NOS (mNOS); and endothelial NOS (eNOS).
eNOS is a calcium/calmodulin dependent enzyme which undergoes several post translational modifications, including acylation with myristate and palmitate, and phosphorylation on numerous residues. As with the other members of the NOS family, eNOS derives the diffusible multifunctional second messenger NO from L arginine through a series of reactions in which L citrulline is an intermediate. eNOS plays an important role in controlling vascular tone, platelet aggregation, and cardiac myocyte function.

Immunoblot Analysis: Whole cell lysates of untreated (Lane 1) and pervanadate-treated (Lane 2) bend 3 cells were separated by SDS-PAGE and blotted to a PVDF membrane. The immunoblot was probed with mab eNOS Antibody Cat.-No AM00098PU-N (Clone 15E2) at 1 μg/ml for 1h at 15-22°C and developed by ECL (exposure time: 3min). Mab eNOS-15E2 recognizes eNOS pSer1177 at 140 kDa.

Proximity Ligation Analysis (PLA) of protein-protein interactions between AKT1 and NOS3 HeLa cells were stained with anti-AKT1 rabbit purified polyclonal 1:1200 and anti-NOS3 mouse monoclonal antibody 1:50. IHC : Human placenta tissue stained with NOS3 polyclonal antibody ( Cat # PAB12680 ) at 1 : 50 for 10 min at RT. Staining of formalin-fixed tissue requires boiling tissue sections in 10 mM Citrate Buffer, pH 6.0 for 10 min followed by cooling at RT for 20 min.

Immunofluorescent analysis of eNOS (pS1177) staining in HuvEc cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).

The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen.
