Background of OAT antibody
OAT is a key enzyme in the pathway that converts arginine and ornithine into the major excitatory and inhibitory neurotransmitters glutamate and GABA. Mutations of this enzyme cause the autosomal recessive eye disease Gyrate Atrophy. This gene encodes the mitochondrial enzyme ornithine aminotransferase, which is a key enzyme is the pathway that converts arginine and ornithine into the major excitatory and inhibitory neurotransmitters glutamate and GABA. Mutations that result in a deficiency of this enzyme cause the autosomal recessive eye disease Gyrate Atrophy. Western blot analysis
The Cell and tissue lysates (40ug) were resolved by SDS-PAGE, transferred to PVDF membrane and probed with anti-human OAT antibody (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL detection system.
Lane 1: OAT recombinant protein (20ng).
Lane 2: 293T cell lysate.
Lane 3: Brain tissue lysate.

Flow cytometry analysis of OAT in Hep3B cell line, staining at 2-5 µg for 1x106 cells (red line). The secondary antibody used goat anti-mouse IgG Alexa fluor 488 conjugate. Isotype control antibody was mouse IgG (black line).

Immunofluorescent analysis of OAT staining in Raw264.7 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
