Background of OS9 antibody
OS9 (protein OS-9 or amplified in osteosarcoma 9) is a lectin/glycoprotein that functions in ER quality control and ER-associated degradation (ERAD pathway) of newly synthesized proteins. TRPV4 is its major target and OS9 also interact with GRP94, DERL2, HSP90B1, CREB3, ERLEC1, HSPA5 etc., forms a part of HRD1 ubiquitin ligase complex (SYVN1/HRD1 and SEL1L) which is involved in retrotranslocation of misfolded proteins from ER to cytosol for ubiquitin dependent degradation. OS9 is essential for recognizition of termilly misfolded non-glycosylated proteins or improperly folded glycoproteins, binding with them for their retention in ER, and transferring them to ubiquitition machinery for degradation. Moreover, it has also been shown to specifically protect some proteins from degradation process before maturation, ensuring their biological functiolity. Because OS9 is implicated in protein quality control, it is closely related to ER stress, and gets up-regulated in response to ER stress. Recently, it has been shown to highly express in adipocytes wherein it regulated lipid storage, and its overexpression is also observed in sarcomas especially in osteosarcoma OSA-CL and rhabdomyosarcoma RH30 cell lines.
