Background of Phospholamban antibody
Phospholamban (PLB/PLN) is a 52 amino acid phosphoprotein which regulates the calcium pump of cardiac sarcoplasmic reticulum (SR). PLB is an oligomer of five identical subunits each having a cytoplasmic and transmembrane domain. The cytoplasmic domain (residues 1 to 25) contains cAMP-dependent protein kinase phosphorylation sites and is highly basic and readily cleaved by proteases; whereas the transmembrane domain (residues 25 to 52) is mostly hydrophobic, protease resistant and stabilizes the pentamer. PLB is an inhibitor of cardiac muscle sarcoplasmic reticulum Ca(2+)-ATPase in the unphosphorylated state, but inhibition is relieved upon phosphorylation of the protein. The subsequent activation of the Ca(2+) pump leads to enhanced muscle relaxation rates, thereby contributing to the inotropic response elicited in heart by beta-agonists. Mutations in this gene are a cause of inherited human dilated cardiomyopathy with refractory congestive heart failure.
