Background of PLK1 antibody
At least four distinct polo-like kises exist in mammalian cells: PLK1, PLK2, PLK3, and PLK4/SAK . PLK1 apparently plays many roles during mitosis, particularly in regulating mitotic entry and exit. The mitosis promoting factor (MPF), cdc2/cyclin B1, is activated by dephosphorylation of cdc2 (Thr14/Tyr15) by cdc25C. PLK1 phosphorylates cdc25C at Ser198 and cyclin B1 at Ser133 causing translocation of these proteins from the cytoplasm to the nucleus. PLK1 phosphorylation of Myt1 at Ser426 and Thr495 has been proposed to ictivate Myt1, one of the kises known to phosphorylate cdc2 at Thr14/Tyr15. Polo-like kises also phosphorylate the cohesin subunit SCC1, causing cohesin displacement from chromosome arms that allow for proper cohesin localization to centromeres. Mitotic exit requires activation of the aphase promoting complex (APC), a ubiquitin ligase responsible for removal of cohesin at centromeres, and degradation of securin, cyclin A, cyclin B1, Aurora A, and cdc20. PLK1 phosphorylation of the APC subunits Apc1, cdc16, and cdc27 has been demonstrated in vitro and has been proposed as a mechanism by which mitotic exit is regulated.Substitution of Thr210 with Asp has been reported to elevate PLK1 kise activity and delay/arrest cells in mitosis, while a Ser137Asp substitution leads to S-phase arrest. In addition, while D damage has been found to inhibit PLK1 kise activity, the Thr210Asp mutant is resistant to this inhibition. PLK1 has been reported to be phosphorylated in vivo at Ser137 and Thr210 in mitosis; D damage prevents phosphorylation at these sites.
Proximity Ligation Analysis (PLA) of protein-protein interactions between CCNB1 and PLK1. Mahlavu cells were stained with anti-CCNB1 rabbit purified polyclonal 1:1200 and anti-PLK1 mouse monoclonal antibody 1:50. Western blot analysis is shown to detect endogenous and recombinant protein present in HeLa cell lysates transfected with various PLK1 mutation constructs.Blots were reacted with PLK1 (phospho T210) polyclonal antibody ( Cat # PAB10069, panel A ) and pan reactive anti-PLK1 ( panel B ).Transfected cells were treated with 1 µM nocodazole followed by cell collection, lysate preparation, SDS-PAGE and western blotting.Using a 1:1000 dilution, PLK1 (phospho T210) polyclonal antibody ( Cat # PAB10069 ) detects a single band corresponding to endogenousplk-1, but does not detect recombinant forms of the protein presumably because of a lack of phosphorylation in thesemutants.Personal communication Hai Jiang, Northwestern Univ.
Western blot of rat synaptic membrane showing specific immunolabeling of the ~66 k Polo-Like Kinase protein phosphorylated at Thr210 (control). The phosphospecificity of this abeling is shown in the second lane (lambda-phosphatase: l-Ptase). The blot is identical to the control except that it was incubated in l-Ptase (1200 units for 30 min) before being exposed to the Polo-Like Kinase (phospho T210) polyclonal antibody ( Cat # PAB9614 ). The mmunolabeling is completely eliminated by treatment with l-Ptase.

Staining of HeLa nuclear lysate using PLK1 ANtibody Cat.-No AP32818PU-N at 0.3 µg/ml (35µg protein in RIPA buffer). Primary incubation was 1 hour. Detected by chemiluminescence.
Hela cell extract (Lane 1) or NIH3T3 cell extract (Lane 2) was resolved by electrophoresis, transferred to nitrocellulose and probed with monoclonal anti-Plk-1 (Clone 3F8) antibody. Proteins were visualized using a goat anti-mouse secondary conjugated to HRP and a chemiluminescence detection system.

Immunohistochemical analysis of PLK1 (pT210) staining in human lung cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

HEK293T cells were transfected with the pCMV6-ENTRY control (Left lane) or pCMV6-ENTRY PLK1 (RC201795, Right lane) cDNA for 48 hrs and lysed. Equivalent amounts of cell lysates (5 ug per lane) were separated by SDS-PAGE and immunoblotted with anti-PLK1.
Immunoprecipitation(IP) of PLK1 by using TrueMab monoclonal anti-PLK1 antibodies (Negative control: IP without adding anti-PLK1 antibody.). For each experiment, 500ul of DDK tagged PLK1 overexpression lysates (at 1:5 dilution with HEK293T lysate), 2ug of anti-PLK1 antibody and 20ul (0.1mg) of goat anti-mouse conjugated magnetic beads were mixed and incubated overnight. After extensive wash to remove any non-specific binding, the immuno-precipitated products were analyzed with rabbit anti-DDK polyclonal antibody.
HEK293T cells transfected with either RC201795 overexpress plasmid(Red) or empty vector control plasmid(Blue) were immunostained by anti-PLK1 antibody(TA500396), and then analyzed by flow cytometry.

Formalin-fixed and paraffin-embedded human cancer tissue reacted with the primary antibody, which was peroxidase-conjugated to the secondary antibody, followed by AEC staining.
Western blot analysis of anti-PLK Pab in A375 cell lysate. PLK (arrow) was detected using purified Pab. Secondary HRP-anti-rabbit was used for signal visualization with chemiluminescence.
WB analysis using Anti-Plk-1 pT210 antibody to detect endogenous protein present in a Mouse A20 whole cell lysate (arrowhead). Comparison to a molecular weight marker indicates a band of ~68 kDa corresponding to Plk-1 protein. It is suggested to use a nuclear extract from synchronized cells to greatly increase the abundance of this protein in preparations. The blot was incubated with a 1:500 dilution of the antibody at RTfollowed by detection using standard techniques.
WB analysis to detect endogenous and recombinant protein present in HeLa cell lysates transfected with various plk-1 mutation constructs. Blots were reacted with anti-Plk-1 pT210 (panel A) and pan reactive anti-Plk-1 (panel B). Using a 1:1000 dilution, anti-Plk-1 pT210 detects a single band corresponding to endogenous plk-1, but does not detect recombinant forms of the protein presumably because of a lack of phosphorylation in these mutants.

Western blot of rat synaptic membrane showing specific immunolabeling of the ~66 k PLK protein phosphorylated at Thr210 (control). The phosphospecificity of this labeling is shown in the second lane (lambda-phosphatase: λ-Ptase). The blot is identical to the control except that it was incubated in λ-Ptase (1200 units for 30 min) before being exposed to the phospho-Thr210 PLK antibody. The immunolabeling is completely eliminated by treatment with λ-Ptase.
