Background of SUZ12 antibody
A chromosomal aberration involving SUZ12 may be a cause of endometrial stromal tumors. Translocation t (7;17)(p15;q21) with JAZF1 generates the JAZF1-SUZ12 oncogene consisting of the N-terminus part of JAZF1 and the C-terminus part of SUZ12. It is frequently found in all cases of endometrial stromal tumors, except in endometrial stromal sarcomas, where it is rarer.This zinc finger gene has been identified at the breakpoints of a recurrent chromosomal translocation reported in endometrial stromal sarcoma. Recombination of these breakpoints results in the fusion of this gene and JAZF1. The protein encoded by this gene contains a zinc finger domain in the C terminus of the coding region. WB was performed on nuclear extracts from HeLa cells (HeLa NE, 20 ug) using the antibody against Suz12 diluted 1:1,000 in TBS-Tween containing 5% skimmed milk. A molecular weight marker (in kDa) is shown on the right; the location of the protein of interest is indicated on the left.
WB using the antibody against Suz12 diluted 1:1,000 in TBS-Tween containing 5% skimmed milk (lane 2, expected size: 83 kDa). A molecular weight marker is shown in lane 1.
ChIP was performed using 600 ug of chromatin from mouse ES cells and 10 ug of the ab against Suz12. IgG (10 ug) was negative control. The IP'd DNA was analysed by qPCR primers for different positive and negative genes. Image shows the results expressed as a % of input (the relative abundance of target DNA in the immunoprecipitates compared to the input DNA).
