Background of ZIC1 antibody
Zinc finger protein ZIC 1 (ZIC1) is a member of the ZIC family of C2H2-type zinc finger proteins. These proteins are particularly important during embryonic development. ZIC1 is a transcriptiol activator via interaction with GLI1, GLI2, and GLI3. ZIC1 facilitates development and maturation of the cerebellum and spil cord during organogenesis of the CNS. Within the pontine gray nucleus (PGN) ZIC1 plays a role in spatial distribution of mossy fiber neurons. The interaction between Zic and Gli family proteins are significant for molecular antagonistic and synergistic factors in development. Zic has been shown involvement in modulation of the hedgehog sigling pathway. ZIC1 may be found in the nucleus or in the cytoplasm in the presence of MDFIC overexpression. This protein is mainly found within the CNS and has high levels of expression in the cerebellum. In cancerous tissue, the ZIC1 protein is often found in medulloblastomas. ZIC1 has been used as aneuroepithelial marker.

Immunohistochemical analysis of ZIC1/2/3 staining in human brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugad compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
Immunofluorescent analysis of ZIC1/2/3 staining in Jurkat cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 deg C in a hidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark. DAPI was used to stain the cell nuclei (blue).
