Background of PPAR-gamma antibody
Since their discovery in the early 1990's, the peroxisome proliferator activated receptors (PPARs) have attracted significant attention. This is primarily because PPARs serve as receptors for two very important classes of drugs: the hypolipidemic fibrates and the insulin sensitizing thiazolidinediones. Peroxisome proliferators are non-genotoxic carcinogens that are purported to exert their effect on cells through their interaction with members of the nuclear hormone receptor family termed PPARs. Nuclear hormone receptors are ligand-dependent intracellular proteins that stimulate transcription of specific genes by binding to specific DNA sequences following activation by the appropriate ligand. Upon binding fatty acids or hypolipidemic drugs, PPARs form heterodimers with retinoid X receptors (RXRs) and these heterodimers regulate the expression of target genes. There are 3 known subtypes of PPARs: PPAR-alpha, PPAR-delta and PPAR-gamma. The target genes of PPAR-alpha and PPAR-delta are primarily involved in the catabolism of fatty acids. Conversely, PPAR-gamma is activated by peroxisome proliferators such as prostaglandins, leukotrienes and anti-diabetic thiazolidinediones and affects the expression of genes involved in the storage of fatty acids. PPAR-gamma may also be involved in adipocyte differentiation. It has also been shown that PPARs can induce transcription of acyl coenzyme A oxidase and cytochrome P450 through interaction with specific response elements.

Western blot using PPARG polyclonal antibody ( Cat # PAB11248 ) shows detection of PPARG protein in a mouse 3T3 whole cell lysate ( lane 1 arrowhead ).Approximately 20 µg of lysate was loaded onto a 4-20% gradient gel followed by transfer to nitrocellulose.Primary antibody was used at a 1 : 2,000 dilution in 5% BLOTTO PBS solution.The membrane was washed and reacted with a 1 : 10,000 dilution of IRDye.IgGTM800 Conjugated Affinity Purified Goat-anti-Rabbit [H&L] MX10.Molecular weight estimation was made by comparison to prestained MW markers indicated at the left ( lane M ).

ChIP was performed on macrophages derived from mouse bone marrow using the ab against PPARG and optimized PCR primer sets for qPCR. Sheared chromatin from 1 million cells and 1 ug of PPARg antibody were used per ChIP experiment. IgG was used as a negative IP control. Figure 1A: recovery, expressed as the % of input, of the PDK4 PPAR response element (RE). Figure 1B: recovery of the FABP4 Adipo PPAR RE in cells treated with RSG, a very strong activating ligand of PPARG, and in untreated cells.
Determination of the titer To determine the titer, an ELISA was performed using a serial dilution of the antibody against human PPARG. The plates were coated with the peptide used for immunization of the rabbit. By plotting the absorbance against the antibody dilution (Figure 2), the titer of the antibody was estimated to be 1:70,250.

Immunohistochemical analysis of PPAR gamma staining in mouse heart formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
